If you are not limiting yourself to audio, certainly the pulse RF transmitters developed bvy the Soviet Union for over the horizon radar purposes that generate megawatts of output would be a good guess. Also, TV stations routinely run megwatt transmitters using Klystrons, a vaccum tube variation. If you are talking about the largest vacuum tube audio amp currently made, then my guess is the Shoreline 800 hi-fi amplifier.
