The tonal considerations have already been covered pretty thouroughly by the other posters here so I won't rehash what's already been said. When I designed the enclosure for my amp, if you even want to call it designing, was really pretty simple. I build the chassis around the circuit that and then the enclosure around the chassis. Once the entire system is built the sound of your amp may change dramatically from what you started out with. At this point you'll have to tweak your circuit a little to compensate for this. I somewhat think that this is what Marshall and Fender did with their designs. The principle behind a speaker cab used in a stereo is to not allow under any circumstances the enclosure to color the sound of what's being reproduced by your stereo. They're shooting for a flat response across the entire frequency spectrum. Cabs used in ginstrument amps on the other hand are an integral part of the total sound, so you want to take this into consideration. You may actually want a little more bass or mids or whatever.
