Try to change the tube, with a known working tube and see what happens. If the tube is glowing again (switch of the power immediately), it might be an open grid1 resistor (or a bad solder joint), or it could be worse, a shorted Output Transformer winding (I'll hope not).  
If not then it is the tube itself
