If your going for a cathode biased amp at first, I would go for the 2 stage/longtail setup for a tweed Princeton type of thing. If you go for the 3 stage/split load setup your getting into a more blackface territory with the grids of your 6v6's getting less signal drive....although with the extra gain stage in the preamp (depending on the configuration) you could pickup more gain from the front end going into the splitload????Bruce,Doc,R.G. and Ken will be able to give you way more details on the more complex relationship between the two.  
 
As far as the tone stack goes,(IMHO) I think a mid control would be more useful than a presence control. But I'm a sucker for the single tone or tre/bass stack.....The less complex a tone stack is..... the less db loss you get...this usually = more preamp drive.  
 
It's all a matter of taste.......and that's the fun part.
